The bus departs from Big Bus Tours Stop #1 at the Termini Railway Station at Via Giolitti 32. You should arrive there thirty minutes before your arrival time. You will need to present your booking confirmation, which you can do by either printing it off or showing your mobile device to a staff member before you board the bus. Seats are given on a first-come, first-serve basis, so if you really want to get to sit on top of the bus you should get there early.
Also, if you do want to sit on the top, make sure you dress warmly. This is wintertime, after all.
